N-Acetylcysteine Displaces Glutathionyl-Moieties from Hg2+ and MeHg+ to Form More Hydrophobic Complexes at Near-Physiological Conditions

چکیده

The anthropogenic release of Hg is associated with an increased human exposure risk. Since Hg2+ and MeHg+ have a high affinity for thiols, their interaction L-glutathione (GSH) within mammalian cells fundamentally involved in toxicological chemistry excretion. To gain insight into the these mercurials multiple small molecular weight we investigated competitive interactions GSH N-acetylcysteine (NAC) at near-physiological conditions, using liquid chromatographic approach. This approach injection each mercurial onto reversed-phase (RP)-HPLC column (37 °C) PBS buffer mobile phase containing 5.0 mM to simulate cytosolic conditions being detected effluent by inductively coupled plasma atomic emission spectrometer (ICP-AES). When was amended up 10 NAC, gradually increasing retention times both were observed. explain this behavior, experiment NAC replicated 50 Tris (pH 7.4), Hg-containing fractions analyzed electrospray ionization mass spectrometry. results revealed presence Hg(GS)(NAC) Hg(NAC)2 MeHg(GS) MeHg(NAC) MeHg+, which suggests that coordination/displacement GS-moieties from more hydrophobic can behavior. biotransformations observed they are relevance as provide biomolecular explanation some obtained when animals administered NAC.

ASH1 mRNP-core factors form stable complexes in absence of cargo RNA at physiological conditions

Asymmetric ASH1 mRNA transport during mitosis of budding yeast constitutes one of the best-studied examples of mRNA localization. Recently, 2 studies used in vitro motility assays to prove that motile ASH1 mRNA-transport complexes can be reconstituted entirely from recombinant factors. Both studies, however, differed in their conclusions on whether cargo RNA itself is required for particle asse...

N-Acetylcysteine Fails to Protect Rats from Acrylamide Neurotoxicity

Acrylamide monomer can impair the central and peripheral nervous system in humans and experimental animals. Although the mechanism of the toxicity has not been clarified, acrylamide has been shown to have a high reactivity with non-protein sulfhydryls. N-Acetylcysteine (NAC), a thiol-containing compound, has been used against acetaminophen overdose due to its potential to stimulate the synthesi...

The hepatoprotective and antioxidant effects of Curcumin and N-acetylcysteine in rats exposed to arsenic

Introduction: Arsenic is a highly toxic element that is widely distributed in environment. Antioxidants depletion and oxidative stress is now considered as one of the possible mechanisms of arsenic-induced toxicity. N-acetylcysteine (NAC) and Curcumin (Cur) are potential antioxidants that can compensate the depletion of antioxidants. This study aimed to compare the hepatoprotective effect of Cu...

Hybridization and dissociation rates of phosphodiester or modified oligodeoxynucleotides with RNA at near-physiological conditions.

We have used RNase H to study both the rates of oligonucleotide hybridization and dissociation at near-physiological conditions. We have studied the Effects of oligonucleotide length, mismatch, and chemical modifications on oligonucleotide association and dissociation with RNA. Dissociation results were compared with standard thermal melting curves to compare relative stabilities evaluated by t...
